A partir de 1726, começou a receber configuração urbana, com base na Engenharia Militar, e também uma arquitetura fortemente influenciada pela Maçonaria, com fachadas e colunas das edificações ornadas com símbolos.
Assim como todas as igrejas de Paraty, a Igreja da Santa Rita possui fachada voltada para o mar. Foi aberta ao público em 30 de junho de 1722, tornando-se a mais antiga igreja do local devido às demolições da Capela de São Roque, das antigas Matriz e Igreja de Nossa Senhora do Mamanguá. Existe nesse anexo um poço de água transparente que muitos acreditam ser milagrosa. Nos fundos da igreja está o Museu de Arte Sacra.
